ABSTRACT

Cancer cell membrane (CCM) derived nanotechnology functionalizes nanoparticles (NPs) to recognize homologous cells, exhibiting translational potential in accurate tumor therapy. However, these nanoplatforms are majorly generated from fixed cell lines and are typically evaluated in cell line-derived subcutaneous-xenografts (CDX), ignoring the tumor heterogeneity and differentiation from inter- and intra- individuals and microenvironments between heterotopic- and orthotopic-tumors, limiting the therapeutic efficiency of such nanoplatforms. Herein, various biomimetic nanoplatforms (CCM-modified gold@Carbon, i.e., Au@C-CCM) were fabricated by coating CCMs of head and neck squamous cell carcinoma (HNSCC) cell lines and patient-derived cells on the surface of Au@C NP. The generated Au@C-CCMs were evaluated on corresponding CDX, tongue orthotopic xenograft (TOX), immune-competent primary and distant tumor models, and patient-derived xenograft (PDX) models. The Au@C-CCM generates a photothermal conversion efficiency up to 44.2% for primary HNSCC therapy and induced immunotherapy to inhibit metastasis via photothermal therapy-induced immunogenic cell death. The homologous CCM endowed the nanoplatforms with optimal targeting properties for the highest therapeutic efficiency, far above those with mismatched CCMs, resulting in distinct tumor ablation and tumor growth inhibition in all four models. This work reinforces the feasibility of biomimetic NPs combining modular designed CMs and functional cores for customized treatment of HNSCC, can be further extended to other malignant tumors therapy.

ABSTRACT

【Objective】 To study the incidence and specificity of platelet antibody in blood donors in Suzhou, analyze the distribution characteristics of platelet antibody in blood donors in this area, and explore the significance of platelet antibody detection in blood donors to reduce the adverse reactions toplatelet transfusion in clinical. 【Methods】 Platelet antibody detection was performed in 2178 blood donors in this area by solid-phase immunosorbent assay. The antibody specificity of the positive samples was analyzed by commercial kit, and the anti-CD36 antibody positive samples were further identified by flow cytometry and gene sequencing. 【Results】 Twelve positive samples were detected by platelet antibody screening, with a positive rate of 0.55%(12/2 178), including 5 males (0.33%, 5/2 178)and 7 females(1.06%, 7/2 178). Among the positive samples, anti-HLA-Ⅰ antibody was identified in 2 cases, anti-CD36 antibody in 1 case, and the antibody specificity was not identified in the other 9 cases. In one case, the positive rate of anti-HLA-Ⅰ antibody PRA was 31.31%(31/ 99), which was mainly specific to anti-B15, anti-B35 and anti-B40. The positive rate of anti-HLA-Ⅰ antibody PRA in the other case was 45.45%(45/ 99), which was mainly specific to anti-A2, anti-A11, anti-A24, anti-A29, anti-A33, anti-A66, anti-B15 and anti-B35. The blood donor with anti-CD36 antibody was type I CD36 deficiency, and 329_330delAC mutation occurred in exon 5. 【Conclusion】 Through antibody screening and specificity identification, the positive rate of platelet antibody in females was significantly higher than that in males(P<0.05). In addition to the common anti-HLA-I antibodies, anti-CD36 antibody was also detected in type I CD36 deficient blood donor. Therefore, the detection of platelet antibodies in blood donors is of certain clinical significance to reduce the adverse reactions to blood transfusion caused by antibodies in platelet products.

【Objective】 To investigate the expression of CD36 antigen in Suzhou area, analyze the type of antigen deficiency and gene mutation, so as to provide references for the establishment of CD36 negative donor registry in Suzhou. 【Methods】 Anticoagulant whole blood samples (805 cases) were randomly collected from healthy blood donors in Suzhou Blood Center. The expression of CD36 antigen on platelet and monocyte was analyzed by flow cytometry to determine the type of CD36 deficiency. The gene mutation type of platelet CD36 antigen-deficient was performed by genomic DNA sequencing. 【Results】 The CD36 deficiency frequency on platelet was 2.48% (20/805), among which TypeⅠ(lacking CD36 expression both on platelet and monocyte) and TypeⅡ(lacking CD36 expression on platelet only) CD36 deficiency accounted for 10% (2/20) and 90% (18/20), respectively. CD36 gene mutations were found in 10 samples, including 3 cases of 329_330 delAC, 1 case of 1228_1239 delATTGTGCCTATT and 2 cases of 1163 A>T; 1 case of 329_330 delAC+ 1172_1183 delTATTGGTCAAGC and 287 G>C+ 329_330 delAC heterozygous mutation. In addition, 1 case of 745 A>G and 1 case of 806 C>T mutations were novel, and not yet reported. 【Conclusion】 Results showed that the frequency of CD36 antigen deficiency in Suzhou were similar to that reported in southern China, but the mutation sites were slightly different. The establishment of CD36 negative platelet registry could provide negative platelets for patients with transfusion reactions caused by anti-CD36 antibody and improve the effect of clinical platelet transfusion.

ABSTRACT

Yuanhuacine is a daphne-type diterpene isolated from Daphne genkwa. It is also isolated from the plant of Daphne and Euphorbia. With the further research on yuanhuacine, it has been found that yuanhuacine has strong anti-tumor activity, and it also exhibits significant anti-virus, anti-inflammatory, neuroprotective and anti-fertility activities. Yuanhuacine can enter various tissues with the blood, and pass through the blood-brain barrier. The bioavailability of yuanhuacine is good, which has potential therapeutic significance for the lesions of all tissues. However, the researches on yuanhuacine mainly focus on the preclinical research and yuanhuacine is only used to induce labor clinically. The purpose of this study is to systematically summarize and analyze the pharmacological activity, drug metabolism in vivo, and clinical application of yuanhuacine, so as to provide a theoretical basis for its development and application.

Objective To explore the effect of applying surface neuromuscular electrical stimulation (NMES) on the functioning of the upper esophageal sphincter (UES) using the high-resolution solid-state manometry.Methods Seventeen healthy volunteers were selected and given sham stimulation (SS),superior and inferior hyoid stimulation (SIHS),superior hyoid stimulation (SHS) or inferior hyoid stimulation (IHS) at a frequency of 80 Hz and the maximum tolerated level before swallowing.Any changes in the kinematics or biomechanics during swallowing and at rest were recorded using high-resolution solid-state manometry.One-factor repeated analysis of the measurement variance was used.Results A significant decrease in the duration of UES relaxation was observed in response to the electrical stimulation (compared with the sham stimulation).The UES's residual pressure showed a rising trend during electrical stimulation,especially in response to IHS,but the difference compared with SS was not significant.The UES's resting pressure during SIHS increased significantly compared with SS,but there was no significant difference between SHS and IHS in this respect.Conclusions Surface neuromuscular electrical stimulation applied to the neck can immediately change the functioning of the UES.NMES at the maximum tolerated intensity can reduce the duration of UES relaxation,which is important for dysphagia therapy and research.

ABSTRACT

Objective:To investigate the relative expression of matrix metalloproteinases-2, 3 (MMP-2, 3) and tissue inhibitors of metalloproteinases-1, 2 (Tissue Inhibitor of Metalloproteinase-1, 2, TIMP-1, 2) in ovarian cancer cell line A2780 under different pressures controlled by carbon dioxide (CO2) changes at different time points and their meaning.Methods:Cell model was established by culture and grouping of A2780 ovarian cancer cell line in vitro.By simulation of laparoscopic CO2pneumoperitoneum environment under different pressure in vitro the pressure of carbon dioxide is controlled at 0 mmHg, 7 mmHg and 15 mmHg respectirely, the cells were collected at 0, 12, 24 and 48 hours after the CO2pressure was controlled for 3 hours.Total RNA was extracted and RT-qPCR was performed to detect the relative expression of MMP-2, 3, TIMP-1 and 2 mRNA in each group.Results:After CO2pressure control, the relative expression of MMP-2 mRNA in each sample under the same pressure decreased with the duration of culture.After CO2pressure control, the relative expression of MMP-2 in ovarian cancer cells decreased with the increase of CO2pressure at the same time point.After CO2pressure control, the relative expression of MMP-3 mRNA in each sample underthe same pressure decreased with the culture time.After CO2pressure control, the relative expression of MMP-3 in ovarian cancer cells decreased with the increase of CO2pressure at the same time point.There was no significant difference in the relative expression of TIMP-1 and TIMP-2 mRNA between the control group and the experimental group at different time points orunder different CO2pressures (P>0.05).Conclusions:The simulated CO2pneumoperitoneum environment could inhibit the invasive ability of ovarian cancer cell line A2780.The relative expression of MMP-2 and MMP-3 decreased astime and pressure increasing.The simulated CO2pneumoperitoneum environment had no significant effect on the expression of A2780 TIMP-1 and 2 mRNA in ovarian cancer cell line A2780.

ABSTRACT

Pre-eclampsia (PE) is a pregnancy disorder that is characterised by severe hypertension and increased risks of foetal and maternal mortality. The aetiology of PE not completely understood; however, maternal nutrition and oxidative stress play important roles in the development of hypertension. The treatment options for PE are currently limited to anti-hypertensive drugs. Punicalagin, a polyphenol present in pomegranate juice, has a range of bioactive properties. The effects of supplementation with punicalagin on angiogenesis and oxidative stress in pregnant rats with induced hypertension were investigated. The pregnant rats were randomly divided into five experimental groups (n=12 per group). Hypertension was induced using an oral dose of NG-nitro-L-arginine methyl ester (L-NAME, 50 mg/kg/day) on days 14–19 of pregnancy. Punicalagin (25, 50 or 100 mg/kg) was given orally on days 14–21 of pregnancy. Punicalagin treatment at the tested doses significantly reduced diastolic, systolic, and mean arterial blood pressure in L-NAME treated rats from day 14. Punicalagin also restored angiogenic balance by increasing the expression of vascular endothelial growth factor and downregulating vascular endothelial growth factor receptor-1/fms-like tyrosine kinase-1. Punicalagin, significantly increased the placental nitric oxide levels as compared to PE group. The increased levels of oxidative stress in rats with PE were markedly decreased by treatment with punicalagin. Punicalagin at the tested doses markedly (p < 0.05) enhanced the placental antioxidant capacity in L-NAME-treated rats. The raised catalase activity observed following L-NAME induction was significantly (p < 0.05) and restored to normal activity levels in punicalagin treatment. Further, 100 mg dose of punicalagin exhibited higher protective effects as compared to lower doses of 25 and 50 mg. This study shows that supplementation with punicalagin decreased blood pressure and oxidative stress and restored angiogenic balance in pregnant rats with induced PE.

ABSTRACT

Objective:To compare the difference of serum levels of M-phospholipaseA2 receptor ( PLA2R) antibodies in patients with idiopathic membranous nephropathy(IMN) detected by two different methods and evaluate the diagnostic value of two methods. Methods:Patients diagnosed as membranous nephropathy and other diseases with biopsy-proven from december 2014 to october 2015 in Shengjing Hospital of China Medical University were enrolled and devided into IMN group and non-IMN group. The serum levels of anti-PLA2R antibody were detected by both indirect immunofluorescence assay(IFA) and enzyme linked immunosorbent assay(ELISA). Results:The sensitivity of IFA and ELISA in IMN were 71. 3% and 68. 5%,and the specificities of two methods were the same as 100%. The area under ROC curves of anti-PLA2R antibody for IMN diagnosis were 0. 860 and 0. 839. The diagnostic value of IFA and ELISA was no statistically significant differences in IMN ( P>0. 05 ) , and the consistency of two methods was better (κ=0. 876). The IMN patients of positive anti-PLA2R antibody be susceptible to the low level of serum albumin (P<0. 05). The higher levels of PLA2R antibody were linked with the worse hypoproteinemia and the higher rate of nephrotic-range proteinuria in IMN patients. Conclusion:Two methods of detecting sera PLA2R antibody have higher sensitivity and specificity,so the sera anti-PLA2R antibody was a better biomarker in the diagnosis of idiopathic membranous nephropathy.

ABSTRACT

Objective To monitor the immediate effect of Passi-Muir speaking valve (PMV) on patients with tracheostomy after acquired brain damage.Methods Twelve patients with tracheostomy after acquired brain damage were recruited.All of them underwent the high resolution manometry to measure the pressure in pharynx and upper esophagus during swallowing before and immediately after wearing PMV.The parameters including the pharyngeal peak pressure,increasing rate of the pharyngeal pressure,pharyngeal pressure duration,upper esophageal sphincter (UES) residual pressure and UES relaxation duration were recorded.Results No significant differences were found in all the measurements before and immediately after wearing PMV (P>0.05).Conclusion PMV has no instant impact on the swallowing function of patients with tracheostomy after acquired brain damage.

ABSTRACT

Objective To explore the effect of the modified balloon dilatation on excitability of the submental swallowing muscle and its relationship with swallowing function of dysphagic patients with brainstem stroke.Methods Thirty unilateral brainstem stroke patients with upper esophageal sphincter (UES) dysfunction were recruited and randomly divided into a dilatation group and a control group,each of 15.The control group was given the routine swallowing therapy twice daily,while the dilatation group was provided one routine swallowing treatment plus a modified balloon dilatation daily.The treatment lasted 3 weeks,30min per day and 5 days per week.Both groups were evaluated using the bilateral submental motor evoked potentials (SMEPs),penetration-aspiration scale (PAS) and functional oral intake scale (FOIS) before and after treatment.Results Repeated analysis of variance showed that there was significant interaction effect of group,time on SMEPs.The MEPs of the affected submental muscle increased significantly in both groups after treatment (P<0.01),but only the amplitude of SMEP of the affected submental muscles in the dilatation group was higher than that of the control group after treatment (P<0.05).In the dilatation group,the average FOIS score increased significantly (P<0.01),while the average PAS score decreased significantly after the treatment (P<0.05).An increase of amplitude of the affected SMEP was strongly correlated with the improvement in the average PAS score (P<0.01,r=0.815),but weakly correlated with the average FOIS score (P =0.02,r =0.43).Conclusion The modified balloon dilatation therapy can increase the excitability of the cortical-affected lateral brainstem-swallowing muscle pathway in patients with unilateral brainstem stroke,which might be more correlated with improvement in swallowing safety.

Objective To analyze the associations of the interleukin-10 (IL-10) promoter-1082G/A and-819C/T single nucleotide polymorphism (SNP) and serum level of IL-10 with intracranial aneurysm (IAs). Methods The polymerase chain reaction (PCR) and DNA direct sequencing methods were used to detect IL-10 gene promoter district two SNP site,-1082G/A and-819C/T genotype frequency and allele frequency in 206 patients with IAs and 187 controls. Chi-square test was used to analyze differences between two groups. The serum level of IL-10 was analyzed by ELISA, and t-test was used to analyze significant differences between two groups. Results There were significant differences in genotypes of GG and GA+AA, as well as the alleles G and A, in-1082G/A locus between IAs group and control group (P<0.01). There were higher frequencies of genotype GA+AA and the allele A in IAs group than those in control group (P<0.01). There was higher risk of suffering IAs in patients with genotype GA+AA (OR=4.137, 95%CI:2.476-6.914) and the allele A (OR=3.368, 95%CI:2.476-4.583). There were higher frequencies of the genotype CT+TT and the allele T in-819C/T locus in IAs group than those of control group (P<0.01). There was higher risk of suffering IAs in patients with genotype CT+TT (OR=3.393, 95%CI:1.952-5.900) and the allele T (OR=3.764, 95%CI:2.730-5.192). The serum level of IL-10 was significantly lower in IAs group than that of control group (P<0.01). Conclusion The IL-10 promoter SNP influences the expression of IL-10. IL-10 promoter-1082G/A and-819C/T polymorphisms are correlated with the formation of IAs.

ABSTRACT

Objective To investigate the relationship between serum cystain C,urinary neutrophil gelatinase?associated lipocalin(NGAL),uri?nary kidney injury molecule?1(KIM?1)and preeclampsia,and explore the value in predicting preeclampsia and early kidney damage. Methods A total of 30 cases of pregnant women with severe preeclampsia,30 cases of mild preeclampsia and 60 cases of healthy pregnant women admitted in the Second Hospital of Jilin University from October 2014 to July 2015 were enrolled for the study. ELISA was performed to check urinary KIM?1 and urinary NGAL,and the serum cystatin C was detected by immune turbidity method. Results KIM?1,NGAL,serum cystatin C in the preeclampsia were significantly higher than the normal control group(P<0.05). In addition,with the development of their concentrations,the progression of pre?eclampsia gradually rise;BMI,e?GFR,urine protein/creatinine ratio,KIM?1,serum scystatin C and NGAL are the risk factors for preeclampsia;uKIM?1,serum scystatin C and NGAL and combined predictors of preeclampsia are taken as the early indicators of renal injury,which sensitivity are separately 83.3%,75%,88.9%,96.8%;specificity are respectively 72.7%,70%,75.6%,86.4%. Conclusion KIM?1,serum scystatin C and NGAL may reflect the progression of renal injury in preeclampsia .Combined predictors have a higher predictive value in the early renal injury of the preeclampsia.

Objective To screen genotypes of small body in Tibet minipigs by single nucleotide polymorphism analysis (SNP) of growth hormone (GH) gene.Methods The PCR products are sequenced.According to the results, SNP analysis are applied in GH gene 5′fragment of 108 Tibet minipigs .Results Five mutation points were found by comparison.The polymorphism information content of the T45C locus is highest.The results of growth traits of different genotypes showed that the abdomen of 6~8 month old Tibet minipigs of TC genotype at T 45C mutation site is small; the weight and height of 3~5 month old Tibet minipigs of AA genotype at G 84A mutation site is small; the body length and height of 6~8month old Tibet minipigs of GG genotype at G 93A mutation site is small.Conclusion The AA TC, GG genotypes of T45C, G84A, G93A mutation sites in GH gene may be associated with small body .We also found that genetic mutations always occur in the above sites with high heterozygosity and genetic diversity , which can provide rich material for breeding research .

Objective To analyze temporal and kinematic parameters of video fluoroscopic images of swallowing using a digital acquisition and analysis system and to verify the reliability of this method.Methods Eighteen patients with dysphagia were requested to completed six swallows (3 ml and 5 ml of thin liquid, thick liquid, and paste mixed with 600 kg/m3 barium sulfate suspension) in the natural sitting position.Video fluoroscopy was used to measure the oral transit times, soft palate elevation times, hyoid movement times, laryngeal closure times, cricopharyngeal muscle opening times, hyoid anterior movement (HAM) , hyoid superior movement (HSM) , cricopharyngeal muscle opening diameter and pharyngeal constriction rate.Each was extracted from the videos four times by two raters working separately with an interval of 4 weeks between the sets of evaluations.Results Reliability varied among the different observations.HAM and HSM showed inter-rater reliability between 0.41 and 0.60 and intra-rater reliability between 0.61 and 0.80.The other observations all demonstrated acceptable reliability.Conclusion The self-designed digital acquisition and analysis system tested showed acceptable reliability and could be applied to analyze swallowing function clinically.

Objective To assess the neurophysiological effects of transcranial direct current stimulation (tDCS) of the motor cortex when swallowing.Methods Twenty healthy volunteers had anodal tDCS (a-tDCS), cathodal tDCS (c-tDCS) or sham tDCS applied over the hemisphere with stronger suprahyoid projections, effortful swallowing was performed simultaneously.Suprahyoid motor-evoked potentials (MEPs) on both the stimulated and non-stimulated contra-lateral hemisphere were examined immediately before stimulation and 5,30, 60 and 90 minutes later.The MEPs were normalized and analyzed using two-way repeated measures analysis of variance.Results The tDCS had long-lasting effects on the suprahyoid MEPs bilaterally.There were significant changes in the effect over time.Sham tDCS showed no significant effect.Compared with sham tDCS, a-tDCS significantly increased the excitability of the stimulated hemisphere, but not the non-stimulated projection.Compared with sham tDCS, c-tDCS induced decreased cortical excitability in the stimulated hemisphere but an increase in the non-stimulated projection.Conclusions tDCS during swallowing can alter bilateral swallowing activity in the motor cortex in a polarity-dependent and site-dependent way.A-tDCS enhances the excitability of the stimulated hemisphere while c-tDCS inhibits it ipsilaterally but increases it contralaterally.

Objective To explore the effects of modified balloon dilatation therapy for treating upper esophageal sphincter dysfunction after a brainstem stroke.Methods Ten healthy adult volunteers and 20 dysphagic patients with upper esophageal sphincter dysfunction after a brainstem stroke were recruited.The 20 patients were divided into a balloon dilatation therapy group of ten who were treated with catheter balloon dilatation therapy, and a control group of ten who received conventional therapy.All were given block-designed task fMRI scans guided by a matched visual presentation system before and after the treatment.Results Widespread activation was observed in both hemispheres, including the bilateral cerebral cortex, the brainstem and the cerebellum, but the activated areas were significantly smaller in the stroke patients before treatment.After the treatment, seven patients in the balloon dilatation group were totally orally fed, while only three patients in the control group recovered totally oral intake.After dilatation, significantly more regions were activated, including the anterior cingulate, insula, supplementary motor area, precuneus and the frontal lobe.They were activated with relatively low voxels in the treatment group, while in the control group significant activation was observed only in the precuneus after treatment.Conclusion Modified balloon dilatation therapy can increase activation of the cortex and subcortical structures related to swallowing, promoting better swallowing function.

Objective To amplify the encoding full-length sequence of Tibet mini-pigs myostatin ( MSTN) gene and analysis the sequence by bioinformatics software.Method The RNA of liver tissues from Tibet mini-pig was extracted, and reversely transcribed into cDNA.The gene coding region sequence of myostatin gene was amplified through RT-PCR, and then the purified product of PCR was ligated with a pMD19-T and transferred into the bacterium DH5αfor replication.The positive clones were screened and sequenced. The sequence characters were analyzed by using bioinformatics method and phylogeny evolution tree was constructed with other twelve species.Results The coding redion of MSTN gene was 1128bp, and coded 375 amino acids.The amino acid homology analysis showed that the homology rate of amino acid sequence was 99%.Conclusions Molecular phylogeny evolution indicated that it had a close relation with human, dog, banna minipig, sheep, goat, cattle, horse, chimpanzee, rat, mouse except chick and zebrafish, and the most closely related with banna minipig.

ABSTRACT

By searching the registry information of systematic review regarding acupuncture included before March 31, 2014 in PROSPERO systematic review registry platform, information in the project plan such as care gories of diseases, interventions, research team and the completion status was analyzed to make a comprehensive understanding on registry status of acupuncture systematic review in this platform. As a result, a total of 52 project plans was finally included. The health problem concerned was mainly painrelated diseases, and the interventions were mostly simple acupuncture or combination of acupuncture and moxibustion. The registered plan participated with Chinese team appeared comparatively late, which was featured with fewer independent projects and concentrated research organization, so its scientific research advantage in acupuncture did not present. In conclusion, it is believed that the consciousness on systematic review registry in domestic researchers needs to be improved, and researchers might take good advantages of the PROSPERO platform in the future.

ABSTRACT

Objective To investigate the significance of galactose galectin-3(Galectin-3)and matrix metalloproteinase-2(MMP-2)expression in gliomas patients and thire role in process of gliomas' malignancy development. Methods Immunohistochemistry was used to detect Galectin-3 and MMP-2 protein expression in 5 normal brain tissue and 40 patients with different grade gliomas. According to positive cells number of Galectin-3,MMP-2 in tumor cells under a microscope,to determine the expression,and the positive index(LI)which came from the percentage of the positive cell number out of the total cell number was to expressed the number of positive cell. Results Galectin-3 and MMP-2 protein expression in normal brain tissue were negative. In glioma tissues,Galectin-3 was mainly expressed in the cytoplasm and membrane of tumor cells. In 23 glioma tissue withⅠor Ⅱ grade,9 cases(39. 13% )was positive and the LI values was(5. 65 ± 3. 47)% in terms of Galectin-3 expression. In Ⅲ,Ⅳ grade glioma specimens,the positive rate of Galectin-3 expression was 76. 47%(13 / 17), and LI value was(27. 88 ± 22. 13)% . The difference of Galectin-3 expression and LI value were significant between specimens with Ⅰ,Ⅱ grade and Ⅲ,Ⅳ significant( χ2 = 4. 101,t = 4. 105;P < 0. 05). In human gliomas,MMP-2 expression protein was mainly expressed in tumor cells and vascular basement membrane of the endothelial cell cytoplasm. In 23 glioma tissue with Ⅰor Ⅱ grade,9 cases(39. 13% )was positive and the LI values was(5. 91 ± 4. 78)% in terms of MMP-2 expression. In Ⅲ,Ⅳ grade glioma specimens,the positive rate of MMP-2 expression was 88. 24%(15 / 17),and LI value was(30. 06 ± 22. 94)% . The difference of MMP-2 expression was significant between specimens with Ⅰ,Ⅱ grade or Ⅲ,Ⅳ grade( χ2 = 7. 882,t = 4. 271;P< 0. 05). The linear correlation analysis showed that there was positively correlation between Galectin-3 and MMP-2 positive cells(r = 0. 800,P < 0. 05). Conclusion Galectin-3 and MMP-2 protein expression in Ⅰ,Ⅱgrade gliomas is significantly lower than those inⅢ,Ⅳ grade glioma,and they are positively related with the progress of malignant gliomas. Galectin-3 and MMP-2 protein can be used to evaluate or judge the malignant stage of human brain glioma.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the injuries of intestinal mitochondria induced by different doses of whole-body radiation in Tibet minipigs.</p><p><b>METHODS</b>Eighteen Tibet minipigs were randomized into 5 radiation groups (n=3) and a control group (n=3). The minipigs in the radiation groups were subject to a total body X-ray radiation at 2, 5, 8, 11, or 14 Gy, and 72 h after the exposure, the mRNA expressions of the intestinal mitochondrial genes were examined using RT-PCR. The changes in the respiratory chain complexes I-IV and the respiratory functions of succinate and NADH were assayed, and the intestinal ultrastructures were observed using transmission electron microscopy (TEM) following the exposures.</p><p><b>RESULTS</b>Compared with those in the control group, the expression levels of the related mitochondrial genes, the activities of the respiratory chain complexes and the function of the respiratory chain were significantly lowered in the radiation groups. At the doses below 8 Gy, the exposures caused significant reduction in the measurements as the radiation doses increased, but at higher doses, these measurements showed no further reductions. Ultrastructurally, exposures at 2 and 5 Gy caused mitochondrial expansion and mild reduction of the density, whereas radiation at 8 Gy or greater resulted in vacuolar changes and obvious expansion of the mitochondria with damages of the mitochondrial cristae and membranes.</p><p><b>CONCLUSION</b>Below the doses of 8 Gy, intestinal mitochondrial damages in the minipigs increase with the radiation dose, but at higher doses, the damages do not further increase with the radiation dose.</p>
